Dozens of new obesity drugs are coming: these are the ones to watch
Nature ^ | February 12, 2025 | Elie Dolgin

Next-generation obesity drugs will work differently from Ozempic and Wegovy — aiming to deliver greater weight loss with fewer side effects.

For Kristian Cook, every pizza box he opened was another door closed on the path to overcoming obesity. “I had massive cravings for pizza,” he says. “That was my biggest downfall.”

At 114 kilograms and juggling a daily regimen of medications for high cholesterol, hypertension and gout, the New Zealander resolved to take action. In late 2022, at the age of 46, Cook joined a clinical trial that set out to test a combination of the weight-loss drug semaglutide — better known by its brand names, Ozempic or Wegovy — and an experimental drug designed to preserve muscle while shedding fat.

Muscle loss is a big concern for people on anti-obesity medications such as semaglutide. These ‘GLP-1 agonists’ mimic a natural gut hormone — glucagon-like peptide 1 — to suppress appetite and regulate metabolism. But reducing calories leads to an energy deficit, which the body often makes up for by burning muscle. The experimental drug that Cook received, called bimagrumab, seems to counteract this muscle loss.

It’s one of more than 100 anti-obesity drug candidates that are in various stages of development. The next wave of medications, which are likely to hit pharmacy shelves in the next few years, resemble drugs that are already on the market. But close behind are numerous therapies being developed specifically for their muscle-sparing weight-loss potential. Dozens more are aimed at different biological pathways and could redefine obesity treatment in decades to come.

“We’re working to create the next generation of healthy weight-loss solutions,” says Philip Larsen, who played a key part in the early development of GLP-1 drugs and is now chief executive of SixPeaks Bio, an obesity-focused start-up company in Basel, Switzerland.

I asked AI what percent of your poop is bacteria.

The percentage of bacteria in human feces varies depending on whether we consider wet or dry weight, but it is a significant component in both cases.

For wet feces:

Approximately 25-54% of fecal matter (excluding water) consists of bacterial biomass
Given that about 75% of feces is water, the actual percentage of bacteria in wet feces is lower, roughly 6-13.5% of the total wet weight

For dry feces:

Bacteria comprise a much larger proportion when considering only the dry weight of feces
50-80% of dry fecal matter is bacteria
54.7 ± 1.7% of total fecal solids is bacteria, according to a study of nine healthy subjects on a British-type diet
55% of fecal dry mass is contributed by bacteria

It’s important to note that this bacterial content includes both living and dead bacteria. The amount of bacteria can vary based on factors such as diet, with high-fiber diets associated with certain types of bacteria that influence stool consistency.

‘I asked AI what percent of your poop is bacteria’

If you imply that probiotics are pooped out and must constantly be replenished, that’s precisely the contrary of what is claimed about them. Take for example poop transplantation for example, as used more and more frequently in Australia to treat patients whose microbiome has been wrecked by antibiotics or potent drugs. It’s about real good bugs which settle permanently in the digestive tract and don’t need refeeding.
Really, probiotics are always glorified fermented foods hyped up by pseudo science and nutrition fads. No less, no more.

‘If you fast and eat Keto your body will burn fat, not muscle. Stop spiking your insulin with 3 meals a day and constant snacking’

Correct. The probem with keto is it’s very easy to ‘fall out of the wagon’ especially for people used to constant snacking. Not to mention the problems with oxalates (joint pains and kidney stone risk). With carnivore, none of these inconveniences exist, you can snack as often as you want and intermittent fasting can come naturally (I personnally eat 3 meals per day at fixed hours, never snack but can skip a meal without any craving). That why a lot of ketos become carnivore to be more consistent and have better results.

‘You don’t lose weight to get healthy....you get healthy to lose weight’

You do lose weight to get healthy, that’s what happens with any diet. Anybody who loses weight becomes healthier, that’s a rule without exception.
On the other hand, you don’t need to be healthy to lose weight, all you need is THE proper human diet (carnivore). On the contrary, the more you are sick and obese, the faster you can lose weight and become healthier.

So your slogan is catchy but not realistic, at all.

For Kristian Cook, every pizza box he opened was another door closed on the path to overcoming obesity.

That's his own fault.

Stop ordering pizza and get mental help for WHY you have an addiction.

I've watched a few episodes of *My 600 Pound Life* recently and it's all basically the same story line. Trauma in childhood, over eating to cope, and enablers who keep feeding them and taking care of them, even when they're bedridden.

Some probiotics are resistant to the stomach acid and do make it to the intestine.

It’s a matter of finding out which ones they are and starting with those.

I asked perplexity.AI the question how fast can new good or bad bacteria colonize the gut? This was the answer. I’ve included some related questions if you like me to post any of those.
..........
New bacteria can colonize your gut remarkably quickly, with significant changes occurring within 3-4 days of a major dietary shift3. Some research suggests alterations to gut microbes can happen within days, though long-term benefits may take several months to manifest5.

The speed of colonization can vary based on several factors:
Diet: Dietary changes can rapidly alter gut microbiome composition35.
Individual differences: Each person’s gut microbiome is unique, affecting colonization rates5.

Antibiotic exposure: Antibiotics can influence the colonization of new bacteria, including potentially harmful ones6.

It’s important to note that while rapid changes are possible, maintaining long-term alterations to gut bacteria requires consistent dietary and lifestyle support5. The gut microbiome is dynamic, and it can return to its original composition if beneficial practices are not sustained5.

Factors Affecting Colonization
Oxygen levels: The lower oxygen environment in the colon favors certain types of bacteria8.

Gut motility: The movement of food and waste through the GI tract affects how bacteria settle and distribute8.
Existing microbiome: A diverse, healthy microbiome can resist colonization by harmful bacteria, while an imbalanced microbiome may be more susceptible8.

In conclusion, while the gut microbiome can change rapidly in response to various factors, sustained changes typically require ongoing support through diet and lifestyle choices.

I cycle in and out of carnivore. After about two weeks I crave vegetables so I do a lot of them for a while, then back to meat. I call it bunny/tiger eating.

Most people fail at keto because they never get their fat ratio high enough. A lot of people are very stuck on fat is bad and just don’t put enough olive oil or butter on their food.
