Molecular mechanism of ergothioneine decoded: Mushroom compound may lead to prolonged health

A team has shown that the natural substance ergothioneine improves the health span of aging animals.

Ergothioneine is a natural compound found in certain fungi such as oyster or shiitake mushrooms as well as fermented foods.

The researchers observed that treatment with ergothioneine from young adulthood not only extended the lifespan, but also increased their mobility as well as their resistance to stress and reduced age-related biomarkers.

The project partners were also able to observe these positive effects in mammals. For three weeks, they treated six nine-month-old rats with a daily dose of about 10 milligrams of ergothioneine—roughly the same amount contained in 4.5 grams of dried oyster mushrooms.

Compared to the control group, the rats' endurance did not only improve, but muscle mass, vascularization of the muscle tissue and the number of muscle stem cells also increased. All of which could make the substance interesting in connection with the prevention of sarcopenia.

Based on human and mouse cell cultures, they were able to show that ergothioneine acts as an alternative substrate for the enzyme cystathionine-γ-lyase (CSE). This enzyme plays a central role in the production of the gaseous signaling molecule hydrogen sulfide (H₂S), which protects cells from oxidative stress through the process of persulfidation.

Reduced persulfidation is associated with aging, cardiovascular diseases and neurodegenerative diseases.

The new research results complement previous findings: Ergothioneine boosts the formation of NAD+—a known coenzyme that has a positive effect on the lifespan.

The researchers also tested the effects of a five-day treatment with ergothioneine on a group of young rats. A daily dose of about 10 milligrams of the substance led to a significant increase in endurance, as well as a significantly increased NAD+ level in the blood serum.

"This indicates that ergothioneine influences in a similar way to performance-enhancing agents," says Filipović.

Ergothioneine lasts about three weeks in the body. We mostly get it from mushrooms and can’t make it ourselves. It acts as a backup antioxidant to two antioxidants we used to make a lot of, when younger, known as superoxide dismutase (SOD) and glutathione.

I’m nearly certain it’s in lion’s mane. There’s some in white button mushrooms but more in other types.

We take a daily mix of 0.5-1.0 grams each of 5 different mushrooms powders.

I recall an unverified report that there is at least one place on DNA where ergothioneine “fits” but nothing else.

I just did a search, yes it does. However cooking removes some of the benefits. Article said microwaving was prefered.

We take a mushroom complex mix, daily, too.

I think ours is Swanson and one is by a Texas organic mushroom grower.

Ergothioneine has its own ion channel into cells to deliver the molecule into at least the mitochondria. There is a special place the molecule then sits, which is right next to its energy production step known for generating free radicals.

I use BulkSupplements.com. Another more expensive but maybe more pure source is MicroIngredients.com.

I asked chapGPT for recommended doses and currently take each day Turkey Tail 0.5g, Reishi 0.5g, Lion's Mane 1.0g, and Cordyceps 1.0g. They run about $45/kg but getting 10% off is easy and 20% off on Black Friday. They have a few other mushroom powders I haven’t researched. I’ll buy organic if they have it.

I currently mix 18 supplements and take 6g 3x per day. It reduces the number of pills I was taking, spreads out the doses, costs less than pills, and may improve bioavailability. It does take a while to measure 50 days worth, mix, and then measure doses. I’ve recently learned amber containers are best, but only have dark green for mixing and storage.

I got started because last year I researched mitochondria health and Dr. Mercola recommended 50-75mg of Niacinamide 3x per day. That’s about 1/64 teaspoon. That’s such a little amount I got other supplements to mix it in.
