NAD⁺ restores memory in Alzheimer's disease models by correcting RNA errors

Alzheimer's disease (AD), the leading cause of dementia, affects nearly 40 million individuals globally. Despite extensive research over the past decades, no treatments have been found that can halt or reverse the progression of this devastating disease.

In AD, a major contributor to neuronal dysfunction is the protein tau.

An international team of researchers has reported a new mechanism by which boosting the natural metabolite NAD⁺ can protect the brain from the degeneration associated with AD.

NAD⁺ (nicotinamide adenine dinucleotide, oxidized form) is a vital metabolite involved in energy metabolism and neuronal resilience in the body. It normally declines with age and especially in various neurodegenerative diseases.

"Preliminary studies have shown that supplementation with NAD⁺ precursors, such as nicotinamide riboside (NR) or nicotinamide mononucleotide (NMN), can offer therapeutic benefits in AD animal models and early clinical trials.

The new study reveals that NAD⁺ works through a previously unidentified RNA-splicing pathway. This pathway is regulated by a protein called EVA1C, which plays an essential role in the process of RNA splicing. Its dysregulation is one of the most recently acknowledged risk factors for AD.

The researchers discovered that when NAD⁺ levels are increased, EVA1C helps correct mistakes in RNA splicing. This restoration process improves the function of hundreds of genes, many crucial for brain health, which can help reverse the neurodegenerative damage caused by tau.

This analysis revealed that NAD⁺ promotes a specific form of EVA1C that efficiently binds to essential proteins, which are central to protein folding and clearance. This connection links metabolic homeostasis, RNA splicing processes and protein management, three processes that are critically impaired in AD.

"We propose that maintaining NAD⁺ levels could help preserve neuronal identity and delay cognitive decline, paving the way for combination treatments to enhance RNA splicing," Ai says.

Taking some of either of the following supplements appears to give this benefit. From the write up:

“Preliminary studies have shown that supplementation with NAD⁺ precursors, such as nicotinamide riboside (NR) or nicotinamide mononucleotide (NMN), can offer therapeutic benefits in AD animal models and early clinical trials.”

I take a small amount of nicotinamide riboside, nearly daily. I get it in a powder form and just put it in water.

It does suck in moisture, in that form, however, so I now keep it in the refrigerator.

You have to be very careful with NAD+ directly. Especially when they come in Strawberry flavored chewable pills. Messes with your pancreas, blood sugar and heart rhythm. Stick with the precursors and let your body decide how much NAD+ to make.

Taking 1000mg of nicotinamide presents some risks. I use to take that amount because it relaxes me but it can down regulate sirtuins and screw up your body’s methylation.

When looking up this stuff then I discovered this:

“some newer e-cigarettes and smokeless tobacco products are being marketed with nicotinamide as a substitute for nicotine, sometimes in combination with other substances like 6-methyl nicotine.”

My partner had some memory issues and such. I gave her neurologist and article about high does of Vitamin D3 that showed it might help. He said that was a good idea and to go on and try it. We had some good results from it and she actually got better.

I had her on like 5000 to 10,000 units a day for along time. At a check up her D test was 114 and normal is 80 so I cut her back, but still kept her level at close to 80. She never got sick and when Covid came we were both on the High Vitamin D3 level and we didn’t get Covid either. I keep my level at close to 70.
