Can weekly prednisone treat obesity? (Apparently “yes”)
Medical Xpress / Journal of Experimental Medicine / Journal of Clinical Investigation ^ | Apr. 1, 2022 | Mattia Quattrocelli et al / Isabella M. Salamone et al

Obese mice that were fed a high-fat diet and that received prednisone one time per week had improved exercise endurance, got stronger, increased their lean body mass and lost weight, reports a new study. The mice also had increased muscle metabolism.

The once-weekly prednisone promoted nutrient uptake into the muscles.

Investigators also found these mice had increased adiponectin levels, a fat-derived hormone that appears to play an important role in protecting against diabetes and insulin resistance.

The scientists also showed mice that were already obese from eating a high-fat diet also had benefit after once-weekly prednisone, experiencing increased strength, running capacity and lower blood glucose.

"Daily prednisone is known to promote obesity and even metabolic syndrome—a disorder with elevated blood lipids and blood sugar and weight gain," McNally said. "So, these results, in which we intermittently 'pulse' the animals with once-weekly prednisone, are strikingly different."

In previously published research, McNally's team discovered giving prednisone intermittently was helpful for muscular dystrophy.

The benefits of weekly prednisone are linked to circadian rhythms, reports another new study from Northwestern and University of Cincinnati published last month in Science Advances.

Human cortisol and steroid levels spike early in the morning before you wake up.

"If you don't give the drug at the right time of day, you don't get the response," Quattrocelli said. "In mice, we obtained good effects with intermittent prednisone in muscle mass and function when we dose them at the beginning of their daytime. Mice have a circadian rhythm inverted to us, as they generally sleep during the daytime and are active at night. This could mean that the optimal dosing time for humans during the day could be in the late afternoon/early evening, but this needs to be appropriately tested."

It needs to be specifically timed for when the prednisone is taken, but it appears to be rather helpful.

I was on this steroid for a short while for a sinus infection. Made me feel great while I was on it, but believe it is not good to be on for long durations!

It makes people retain water. Yes, lots of nasty potential side effects. I would not recommend it for weight loss.

This article is about me and I’m taking notice. I wonder what would be a reasonable dose for once/week instead of daily 2.5mg?

I had a very bad case of Polymyalgia rheumatica (Also known as: PMR)

I couldn’t dress myself, every joint in my body was frozen, I couldn’t chew my food.

My Doctor put me on Prednisone and it knocked the PMR out overnight.

The side effects of taking prednisone were; it destroyed my immune systen, and I came down with whooping cough, it also destroyed my A1c diabetic reading.

It took a year to wean me off of prednisone, but my immune system came back, and my PMR is gone.

I also know 3 women who were on prednisone and they all became belligerent and combative, and would break out in tears for no good reason.

Prednisone has a lot of side effects.- Tom

They are saying, “take it in the evening.” Really? I was told to take my daily dose (and for a time it was 100mgs per day) as soon as I could in the AM as it causes insomnia. I think I may have felt it. I had just returned from East Asia when I was first prescribed and it did seem jet lag lasted much longer than previously.

I wonder if this could have a application with respiratory patients. Would a dose once a week be considered continuous use?

It would not be any sort of normal “continuous” use

It appears to be infrequent enough that unusual benefits come from it, despite being once every seven days.

I would think this would be best to take “at the beginning of” your day, which for humans is morning.

It’s odd they are saying it’s possible to be best in the late afternoon.

That’s what I was thinking as well. I still have some. I’m glad I did. I developed shingles while in a remote location and couldn’t get the acyclovir immediately. I began treating with prednisone and benadryl. Finally, after 4 or 5 days I was able to get some acyclovir. The doc that treated me said the combo I took was the only thing, apart from acyclovir, that I could do.

Huh.

Possibly because it reduces inflammation and pain and that leaves you inclined to move more?

Mom-in-law had cancer - prednisone treatment helped with the pain. She was also diabetic and it drove her sugars out of whack. Doctor said her comfort was worth the sugar spike - she didn’t last long.

I was on prednisone for a year..Had to stay away from citrus, sugar, and even food high in sodium . Took Prilosec to protect my stomach lining. Cwey nasty side effects during the initial high dose treatment, among them insomnia, and a horrible empty feeling in my stomach when I woke up
