Scientists led by Dr David Fairlie from the University of Queensland, Australia, have found abnormal amounts of an inflammatory protein called PAR2 in the fat tissues of overweight and obese rats and humans. PAR2 is also increased on the surfaces of human immune cells by common fatty acids in the diet. When obese rats on a diet high in sugar and fat were given a new oral drug that binds to PAR2, the inflammation-causing properties of this protein were blocked, as were other effects of the high-fat and high-sugar diet, including obesity itself.
"This important new finding links obesity and high fat, high sugar diets with changes in immune cells and inflammatory status, highlighting an emerging realization that obesity is an inflammatory disease," Dr Fairlie said. ..... < snip >
..... To make the discovery, the team discovered the first potent, selective and orally bioavailable small molecule antagonists of PAR2 and characterized their functional responses in inflammatory cellular and animal models.
They found that PAR2 expression is increased in vivo in adipose tissue from obese humans and rats, stimulated in vitro in human macrophages by the dietary fatty acid palmitic acid, and inhibited in vivo and in vitro by a PAR2 antagonist. This antagonist was then used as a tool to dissect roles for PAR2 activation in mediating metabolic dysfunction in human monocyte-derived macrophages (HMDM), human and rodent adipocytes, and diet-induced obesity in rats.
Oral treatment of diet-induced obese rats attenuated PAR2 signaling in adipose tissue and inhibited adipose inflammation, insulin resistance, diet-induced obesity and cardiovascular abnormalities.
This is the first report that a PAR2 antagonist improves obesity, glucose homeostasis and obesity-associated chronic inflammation in vivo. ..... < snip >

i just left a message for my doctor about this, asking if he can get rid of blubber now that we know its just an inflammatory reaction?
we will see what he comes up with.
he’s previously told us..that if he knew how to reduce weight, he would treat himself first....doctor heal thyself and all that, ha!

Unfortunately, in today's rigid legal and regulatory environment, doctors may not be [legally] allowed to recommend the best treatment for the patients that they would choose for themselves.
Perhaps some hope for those of us with Crohns or RA.

I just lost 21 pounds in the past 2 months. I eat only meat, green veggies, eggs and cheese. Also nuts and sugar free jello pudding for dessert. No wheat rice potatoes or sugar.

It works quite well but you’re going to have a carb crash, prepare yourself for an episode of overwhelming craving. It hit me after about six months, it was no fun. I’d suggest introducing a very small amount of carbs to prevent this, maybe seasoned roasted potatoes once a week.

If it was sedentary lifestyles and plentiful food alone that caused the obesity epidemic, then it should have taken off in the 1950's.
Instead it took until the late 80s and 90s to really become a problem.
One hypothesis is that a virus spread throughout the population in 1980s-90s that perhaps was ignored because the immediate effects were either limited or similar to common cold/flu viruses.
Yet, this virus was different enough that it caused inflammation/immune response that led directly to obesity or subtle behavioral changes that eventually manifested itself as obesity.
I don’t remember a lot of sedentary lifestyles in the 50s at all. At least in the blue collar ranks, everybody was working pretty hard.
Of course darn near everyone smoked, maybe the nicotine kept everybody slim?
Obesity is a gland problem. So stop eating so many glands.
