No. The President appoints a successor and that successor is confirmed by the Senate. At least that's the way it worked when Gerald Ford became VP.
If Cheney and Dubya went hunting together and shot each other, then Hastert would become the President.

That's how Gerald Ford became Vice President when Agnew resigned.

That is why they call it a Presidential line of Succession. If a VP resigns, a new one is nominated by the President and its then approved by the Senate. If its not approved, the spot stay unfilled and the Speaker than is next line of Succession.

Amendment 25, Sec. 2 states,
Whenever there is s vacancy in the office of the VP, the President shall nominate a VP who shall take the office upon confirmation by a majority vote of both houses of Congress.
