Indiana University School of Medicine researchers have found that niacin limits Alzheimer's disease progression when used in models in the lab, a discovery that could potentially pave the way toward therapeutic approaches to the disease.
The study investigates how niacin modulates microglia response to amyloid plaques in an Alzheimer's disease animal model.
Gary Landreth, Ph.D., Martin Professor of Alzheimer's Research, and Miguel Moutinho, Ph.D., postdoctoral fellow in Anatomy, Cell Biology and Physiology, led the study.
"This study identifies a potential novel therapeutic target for Alzheimer's disease, which can be modulated by FDA-approved drugs," Moutinho said. "The translational potential of this strategy to clinical use is high."
Niacin, which sustains metabolism throughout the body, is mainly obtained through a typical diet; it also can be taken in supplements and cholesterol-lowering drugs. The brain, however, Moutinho found, uses niacin in a different manner.
In the brain, niacin interacts with a highly-selective receptor, HCAR2, present in immune cells physically associated with amyloid plaques. When niacin—used in this project as the FDA-approved Niaspan drug—activates the receptor, it stimulates beneficial actions from these immune cells, Landreth said.
"After the Alzheimer's disease animal models received niacin, they ended up with fewer plaques and they have improved cognition," Landreth said, "and we directly showed that these actions were due to the HCAR2 receptor."
Past epidemiology studies of niacin and Alzheimer's disease showed that people who had higher levels of niacin in their diet had diminished risk of the disease, Landreth said. Niacin is also currently being used in clinical trials in Parkinson's disease and glioblastoma.
To further their research into niacin and the brain, Landreth and Moutinho are collaborating with Jared Brosch, MD, associate professor of clinical neurology, who is applying for a clinical pilot trial to study the affects of niacin and the human brain.

Nuts! You pulled Comment #6 before I had a chance to print it. Could you please repost the link for the book and the name of the author. There was also something with a number of letters named in what you wrote. I was wondering if this was the name of gene I have read that is found in some people with Alzheimers. My husband died of Alzheimers, and I have 2 sons and 4 grandsons. I wanted to help them discover if they have this genetic tendancy All 4 grandsons have been on ADHD or OCD medication in their youth, so I am trying to find connections and possible non-drug treatments.
I have been studying and using supplements for 50 years to prevent or cure various health problems I and those I know well have experienced. In spring 2020 when Fauci answered someone that Vitamins C and D were not necessary to take, I was sick with anger. He, perhaps singlehandedly prevented President Trump from implementing the Zelenko protocol by helping to prove that HCQ (if used without the recommended ZINC) does not significantly improve Covid illness. That anti-science may have killed 1/2 million people these past 2 years. Once HCQ had been vilified, Fauci began touting Remedesivir (at about $3,000 per treatment course, subsidized mostly by our taxes) which has probably cost us many $millions with little result.
The joke by FR890 that they might block niacin is not funny. They have already blocked HCQ, Ivermectin, and possibly NAC. Vitamin C has been marginalized, and now that the link below and others have shown almost no one with OPTIMAL Vitamin D levels ends up in hospitals, that could be next. So far as prophylaxis against Covid and other illness is concerned, I think that getting our people all up to the OPtIMUM level of blood Vitamin D would be the quickest, cheapest, least controversial way to control Covid as a major public health problem.
NOTE: In the US, Kaiser Permanente lists OPTIMUM RANGE of Vitamin D as 50 to 80 ng/mL. The above study uses nmol/L as their blood Vitamin D figure. This is 2.5 times the US measure, so their OPTIMUM of 75 nmol/L is the equivalent of a much lower figure, 30+ ng/mL, than the Kaiser Permanente range. Still not a single one of over 200 they tested as possible trial subjects was OPTIMUM.

I used to take Vitamin E, but switched to tocotrienols, instead. I will have to further consider what to do, because most of the supplements were given once a day, so splitting them up won’t help.

The United States Patent Office delayed issuing a patent on the Wright brothers’ airplane for five years because it broke accepted scientific principles. This is actually true. Dr Hoffer wrote a book or an article on his use of niacin in healing his own mother of alzheimers with niacin.

If concerned about cancer, there are many other substances proved useful, many of which are not drugs with substantial and well-known side effects.
I am already taking statins, have been for 30 years, and am always interested to read of possible beneficial “side effects”.
Should also be informed about negative side effects.
Muscle Pain and Weakness
The most common side effect is muscle pain and weakness, a condition called rhabdomyolysis, most likely due to the depletion of Co-Q10, a nutrient that supports muscle function. Dr. Beatrice Golomb of San Diego, California is currently conducting a series of studies on statin side effects.
Neuropathy
Polyneuropathy, also known as peripheral neuropathy, is characterized by weakness, tingling and pain in the hands and feet, as well as difficulty walking.
Heart Failure
We are currently in the midst of a congestive heart failure epidemic in the United States–while the incidence of heart attack has declined slightly, an increase in the number heart failure cases has outpaced these gains. Deaths attributed to heart failure more than doubled from 1989 to 1997.14 (Statins were first given pre-market approval in 1987.) Interference with production of Co-Q10 by statin drugs is the most likely explanation. The heart is a muscle and it cannot work when deprived of Co-Q10.
Dizziness
Dizziness is commonly associated with statin use, possibly due to blood pressure-lowering effects.
Cognitive Impairment

Cancer
In every study with rodents to date, statins have caused cancer.26 Why have we not seen such a dramatic correlation in human studies? Because cancer takes a long time to develop and most of the statin trials do not go on longer than two or three years. Still, in one trial, the CARE trial, breast cancer rates of those taking a statin went up 1500 percent.27
Pancreatitis

It’s a B vitamin.
It’s a nutraceutical, not a pharmaceutical.
I think I would have experienced negative side effects at some point in the last 30 years if I was going to. My mother also took a statin for all those years until she died at 102 after a fall.
