A new preclinical study has demonstrated how an experimental drug can treat type 2 diabetes by increasing the expression of a recently discovered protein found to influence insulin signaling in cells. Across several mouse studies, the research shows the novel compound improved aspects of metabolic syndrome associated with diabetes.
In the early 2000s, a protein dubbed SWELL1 was identified and it was initially thought to solely play a role in lymphocyte function. Around a decade later, researchers discovered SWELL1 also influences insulin signaling in many tissues across the body. Subsequent study revealed SWELL1 activity is vital for normal insulin secretion from pancreatic cells. And, SWELL1 activity seems to be significantly disrupted in type 2 diabetic patients.
“Since T2D [type 2 diabetes] is characterized by both a loss of insulin sensitivity of target tissues (fat, skeletal muscle, liver) and ultimately, impaired insulin secretion from the pancreatic β-cell, these data raised the question: could impaired SWELL1-mediated signaling contribute to T2D pathogenesis, and if so, could this be corrected pharmacologically to improve systemic glycemia?” the researchers wrote in the new study.
Rajan Sah, senior author on the study, says SWELL1 has a double life, affecting insulin sensitivity in a number of cells across the body, while at the same time influencing insulin secretion from the pancreas. So his team developed a molecule called SN-401 to augment SWELL1 expression in the hopes it could normalize metabolic function in diabetic animals.
“This protein, SWELL1, has a sort of dual personality,” explained Sah. “The compound binds to SWELL1 in a manner that stabilizes the protein complex so as to enhance expression and signaling across multiple tissues, including adipose, skeletal muscle, liver, the inner lining of blood vessels, and pancreatic islet cells. This restores both insulin sensitivity across tissue types and insulin secretion in the pancreas.”

Studies have shown that people without diabetes live an of average eight years longer than people with diabetes.
Now, people with type-2 diabetes who take metformin out live the average of the people who don't have diabetes.

The caveat is “within reason”. Meds all have side effects, some are not worth the side effects. Others are essential, side effects be darned. Sorry that you require so much medication, but if it keeps you going, that’s all that matters.

They've begun a very large study in England to test if the 'metformin effect' affects the telomeres of healthy people.
These people say they will sell metformin W/O a prescription.

I see your type of response often on FR. Your uneducated opinions on disease. Most people like you seem to think diabetics are lazy, fat and eat too much.
My husband developed T2D, not fat, not lazy, eats a normal man’s diet. No family history of T2D. Where or how did he manage to contract this disease? Agent Orange exposure. Sadly, that’s just the beginning.
Now STFU please with your uneducated opinions on other people’s health conditions and causes.
Always lose as much weight as feasible and keep moderately physically and mentally active and cut all extraneous sugars to see if optimal physical condition can alleviate dbt2....then, if not, use the meds...

I've lost 75 pounds so far (got about 25 to go) and it's made a huge difference in my blood sugar management. My last A1c was 6.4 (had been about 7.5).

One side effect from Metformin is that it may degrade your kidneys. I was taking 2000mg a day and had to cut it back to 1000mg a day. Now my kidney function has stabilized and is slowly increasing. It try to limit my carb intake to around 100mg a day.
I am quite educated on the matter, and you the one who sounds silly.
Agent Orange exposure is anomalous for the vast majority of people and the toxic effects on the liver and pancreas well known.
For everyone else, they are a closed thermodynamic system which responds predictably to all those rules about energy in, heat, movement and waste out.
And at least 99.9% of all with T2D have it because of too much of the wrong kinds of food and too little movement.
Ironclad, inescapable rules apply to every person.
Except, of course, those in denial and those who live in never never land.
