Role of pyrethroid derivatives in autophagy and apoptosis crosstalk signaling and potential risk for malignancies (‘Harmless’ insecticides aren’t so harmless)
Medical Xpress / Impact Journals LLC / Oncotarget ^ | Dec. 28, 2022 | Jyothi Puvula et al

Pyrethroids are extensively used insecticides by virtue of insecticidal activity potential in Asia, especially India, and in different nations worldwide to counter mosquitoes and insects for household or agricultural needs. The continuous widespread and uncontrolled use of pyrethroids and its derivatives have influenced multiple deleterious effects resulting in a potential risk factor causing damage to organ systems.

Allethrin and prallethrin are extensively used, yet their influences on human primary cells are very limited or under-reported. The potential mechanisms by which allethrin and prallethrin modulates human primary cells, especially the molecular mechanisms or interconnectivity of autophagy-apoptosis, and their clinical relevance in human subjects or patients are not well defined.

Researchers furnished evidence that both allethrin and prallethrin user samples significantly induced Ccl2 mRNA expression, increased amount of reactive oxygen intermediate, inhibited membrane bound enzymes and altered membrane fluidity.

Pyrethroid derivative users had induced levels of lipid peroxidation and induced binding activities of transcription factors(tfs). Pyrethroid derivatives induced autophagy, elicited intracellular Ca2+ concentration, calcineurin and regulated proapoptotic genes, DAPK1, Bim.

"Our study presumably comprises the initial investigation of a new mechanism of pyrethroid derivatives-moderated programmed cell death in various cell sets or types, like human primary cells where-in this is a late event, is documented," state the researchers.

Hence, the current research-study might be significant in the various pyrethroid derivatives-allied hematological-related cancers and immunosuppressant or auto-immune disorders. In the foremost instance, the researchers present data stating that pyrethroid derivatives induces multiple cell signaling cascades; synchronously effectively impact on the apoptosis, therefore causing hematological tumors and toxic or immune related disorders.

"Overall this current study might facilitate to formulate therapeutics or intervention targets that might serve to decrease the effect or impact of pyrethroids derivatives by targeting the signaling cascade that serves to minimize the modulation of autophagy mediated apoptosis," the researchers conclude.

Autophagy is the self-eating of cells. Apoptosis is the self-destruction of cells. These can be constructive and are normal processes, but you don’t want to encourage healthy cells to do this, especially in organs in which cells don’t properly renew, such as the heart or kidney.

So I use pyrethrin on clothing. Similar issues?

And expect a high-priced patented replacement soon.

I do not find this surprising, but it will certainly not stop me from using my Thermacell devices when camping or whatever.
They do have a warning not to use near food.

There are a lot of similar -metherin pads and heating devices to release this chemical sold in asia.

IIRC SC Johnson is the largest distributor

Rather than a “Natural” product made from daisies and marigolds

“How reliable”

I won’t pay extra.

Some things like apples tend to require pesticides.

Organic does not imply pesticide free.

Nicotine is a pesticide I believe.

I suspect that the danger from various biting insects is greater.

Doesn’t mean we shouldn’t look for even safer compounds, but it is all a risk factor assessment.

I don’t buy insect repellent that has to be “washed off” in the directions.

Pyrethrins are pesticides found naturally in some chrysanthemum flowers. They are a mixture of six chemicals that are toxic to insects. Pyrethrins are commonly used to control mosquitoes, fleas, flies, moths, ants, and many other pests.

Painted daisies are also called pyrethrum, Persian pellitory, or Persian insect flower because they’re insect-repellant. These perennial flowers feature a yellow center surrounded by white, red, magenta, and marigold-colored petals.Sep 9, 2021
