Exploring the brief use of rapamycin treatment in early adulthood to extend lifespan

Imagine you could take a medicine that prevents the decline that come with age and keeps you healthy. The current most promising anti-aging drug is rapamycin, known for its positive effects on life and health span in experimental studies with laboratory animals. To obtain the maximum beneficial effects of the drug, it is often given lifelong. However, even at the low doses used in prevention for age-related decline, negative side effects may occur, and it is always desirable to use the lowest effective dose.

A research group has shown in laboratory animals that brief exposure to rapamycin has the same positive effects as lifelong treatment opening new doors for a potential application in humans.

The scientists have tested different time windows of short-term drug administration in fruit flies and found that a brief window of two weeks of rapamycin treatment in young, adult flies protected them against age-related pathology in the intestine and extended their lives. A corresponding short time window, three months of treatment starting at three months of age in young, adult mice, had similar beneficial effects on the health of the intestine when they were middle-aged.

"These brief drug treatments in early adulthood produced just as strong protection as continuous treatment started at the same time. We also found that the rapamycin treatment had the strongest and best effects when given in early life as compared to middle age. When the flies were treated with rapamycin in late life, on the other hand, it had no effects at all. So, the rapamycin memory is activated primarily in early adulthood," explains Dr. Thomas Leech.

"We have found a way to circumvent the need for chronic, long-term rapamycin intake, so it could be more practical to apply in humans," says Dr. Yu-Xuan Lu.

Look at all the effects of the mTOR system.(see wikepedia, for an example) Ya’ think we’re gonna tease out the mechanism of action soon? There are also other naturally occuring foods which have some mTOR activity. Everyone wants a magic pill....uhhh...uhhh..uhhhh....not so fast....

Cherry the fruit, is supposed to do the same as this drug, without the side affects.

We take 1,800 mg of Glycine, 1,800 mg of NAC, 1,000 mg of Urolithin A, and 1,500 a 2,000 mg of Taurine a day.

Nestle's Celltrient Cellular Strength is what we've been getting for Urolithin A. Amazon has it on subscription at a reasonable price, but there are other places that also carry it, sometimes for rather lower prices.

Thank you. You’re very kind to do all this.

May I ask you about the reasoning behind your dosages?

Nestle’s product is 2 600mg doses of Glycine and NAC combined and taken twice daily for a total of 1200mg each. Why are you taking more?

From the study on Taurine it indicated 3g a day, but you’re taking less. I haven’t seen a dose suggestion regarding Urolithin A. Is that all Nestle offers (2x 500mg/day) or are there studies supporting 1000mg dose/day. If you wouldn’t mind explaining your thinking behind your dosing, I’d appreciate it very much.

We are taking more because it's cheap and the study with 70-80 year olds used nearly 7 grams of each, but didn't try less. We aren't as old, either, so less should be fine.

The study I posted on Taurine did mention 500 mg 3 times a day, but said more would likely be better.

The Urolithin A studies have used either 500 or 1,000 a day. At least one study used both.

I can say the doses my wife and I are taking are working well for us.

The study on the high dose GlyNAC described for the older people involved that they had very few issues, but the maximum time used was 24 weeks and they were under close monitoring.

You should read the studies and determine what you want to do, if anything.

Thanks. I am and I will. I am not relying on you, so I hope it doesn’t seem like that. I respect your insights. I’m taking 3g of Taurine and otherwise following the studies guidelines. I have not added UA, but may. It’s hard to know if the expense is worth it. Do you know of any tests that would clarify the results individually?

It helped human and mouse test subjects, but with the Taurine and GlyNAC, results would be confounded.

Taurine and GlyNAC are a less expensive alternative with some overlapping benefits.
