A young boy died in a trial for Pfizer’s experimental gene therapy for Duchenne muscular dystrophy, the company told patient advocates Tuesday.
The boy was enrolled in Daylight, a trial studying the treatment in boys aged 2 or 3. The boy had received the therapy early last year, Pfizer told the advocates in a note posted online by Parent Project Muscular Dystrophy.
Pfizer said it had not yet determined precisely what happened or whether it was linked to the treatment. Deaths and severe side effects linked to gene therapy have generally happened soon after dosing.

I work in gene therapy. Some work. Others don’t. My take is that the article is irresponsible for suggesting the therapy killed the patient. Sadly, sometimes the patients pass.
Not everything Pfizer does uses mRNA. This therapy uses a viral vector to deliver the correct gene material and it’s an infusion from an IV bag, not a shot.
Because other creatures don’t get human genetic syndromes.
Even if they use a viral vector they are most likely still using the lipid nanoparticle carrier and the n methyl pseudo uridine to make the inserted RNA last longer. Both of these were causing the problems in the mRNA covid vaxx.
It’s DNA not RNA. I see no reason that they would need to use substances developed for RNA when doing DNA gene therapy which has been around a lot longer and has nothing to do with the immune system or vaccination.

It is a fascinating field. They just approved a gene therapy in 2024 for people who have Thalassemia that have to have blood transfusions. I only have the minor version so no transfusions needed.
There is scant info out there but I have seen studies talking about how having even Minor Thalassemia can make you more susceptible to other disorders like Diabetes. Some think it is the location of the gene. There are other theories.
So far no Diabetes for me but a few of other weird and rare disorders.

The article as posted didn’t mention whether it was RNA or DNA. I would imagine they would still use the lipid nanoparticle carrier for both since that was used to assist the RNA/DNA getting past the cell wall and into the ribosomes for RNA or into the nucleus if they used DNA.

I actually looked it up as I have always been interested in all things genetic. They are using a viral vector as the cellular delivery system.
Basically infecting the entire body with a functional gene to counteract the failings of the incomplete damaged gene.
I don’t think the nanoparticles have the right vectoring characteristics for such a systemic whole body treatment.
Gene therapy is a completely different science than vax development.
Don’t we use humanized mice for some human genetic issues?
Humanized mice are mostly just used for testing and initial trials.
