Doubtful. They don’t show Vader ever going down to Tatooine in “A New Hope”, and we know he hates sand, so he probably wasn’t in any mood to visit. I think it was just the Storm Troopers.
Plus, if Vader had gone down to the planet, he probably would have sensed Obiwan was nearby, like he did when Obiwan was on the Death Star. So that also weighs against Vader being responsible.

Luke's uncle was the son of Vadars’ step-father. Would that make him a step-brother?
The movie suggests it was storm troopers who did it after they blasted the Jawa transport with their too precise hits.

Anakin (Darth Vader) was at least peripherally responsible for killing his Uncle Owen. Perhaps it was at that point he learned (through interrogation) that his offspring not only survived childbirth but were twins and had been raised in hiding.
When I see that scene and Luke's resolve to join OB1 because he has nothing left at home and his adoptive parents were killed by the Empire, I often think of the real world situation Einstein was in. Would Germany had won World War II if Einstein hadn't convinced the U.S. to start the Manhattan Project (and thus Germany had been first with the atom bomb)? Would Einstein have written the letter if Germany hadn't done the Holocaust?

I stopped watching after the 2nd or 3rd edition so I have no idea if there’s a definitive answer. We may never know. With the writers and actors on strike the mystery will persist until AI can figure it out.

Luke’s aunt and uncle had the misfortune of getting in the way of the storm troopers who were tracking down R2D2 and CPO3. The storm troopers gunned them down just as they did those Gypsy-type critters who first acquired the robots after they landed on the planet.

First, he’s the step-uncle.
Anikan was born of Shmi Skywalker, a slave, father unknown.
Shmi married Cliegg Lars after Anikan left. Some sandpeople killed Shmi, so Anikan killed them and buried his mother at the Lars’s farm. Cliegg died shortly thereafter, and left the farm to his brother Lars. Kenobi brought the infant Luke to the Lars farm to be raised by his Step-Uncle Owen and his wife, who were unable to have children.
Second, no Anikan/Darth did not kill him.
