INOVIO's Pan-COVID-19 Vaccine Candidate (INO-4802) Induces Broad Immunity Against Major Viral Variants in Preclinical Studies

INOVIO's Pan-SARS-CoV-2 vaccine candidate INO-4802 is not matched to a single variant, unlike other variant vaccines in development; INO-4802 could potentially offer broader immune responses

Phase 1/2 clinical trials with INO-4802 are planned this year

INOVIO (NASDAQ:INO), a biotechnology company focused on bringing to market precisely designed DNA medicines to treat and protect people from infectious diseases, cancer, and HPV-associated diseases, today announced that its next-generation Pan-COVID-19 vaccine candidate, INO-4802, induced potent neutralizing antibodies and T cell responses against the original Wuhan strain as well as against B.1.1.7 (UK variant), B.1.351 (South African variant) and P.1. (Brazilian variant) in preclinical models. These results demonstrate the potential of INOVIO's Pan-COVID-19 vaccine to induce cross-reactive immune responses against current and emerging viral variants as either a first-line vaccine, or potentially as a boost for individuals previously immunized with various Wuhan-matched vaccines.

Dr. Laurent M. Humeau, INOVIO's Chief Scientific Officer, said, "INOVIO is taking a dual-track approach in developing a COVID-19 vaccine because we recognize the need to support both pandemic and endemic considerations. In addition to our work on INO-4800, which we expect to enter a global Phase 3 trial this summer, we are also developing our next-generation Pan-COVID-19 variant vaccine, INO-4802, which is designed to protect against current and potentially future variants of concern. The study data we report today confirms our two-tiered development path is the best strategy for the short- and long-term fight against this virus."

First-generation COVID-19 vaccines were matched against the original Wuhan strain. These vaccines may have reduced efficacy against emerging COVID variants. In response, many of the next-generation vaccines under development are matched to a single variant currently circulating. INOVIO's Pan-COVID-19 vaccine approach is designed to provide cross-strain protection, immune coverage, reduced susceptibility to escape mutants, and non-restricted geographical use against both known and potentially unknown variants.

To create INOVIO’s Pan-COVID-19 vaccine candidate, variant sequences were identified over a four-month period starting in October 2020 from multiple geographic regions (Brazil, Canada, India, Italy, Japan, Nigeria, South Africa, United Kingdom, and the United States). Mutations in the spike sequences were aggregated for each region.

The sequence results from these regions were then further engineered to determine a common set of overlapping mutations from emerging variations in the COVID spike protein sequences to generate INO-4802.

Yup. That’s all I want. A synthetic DNA vaccine.
I am not a virologist but I would guess that there are many antigenic sites on coronavirus variants. Why tinker with dna, rna...when one can kill or attenuate the viruses, break them down, extract and dispose their dna or rna...and give the remaining parts as a vaccine to stimulate an antibody response?

Interesting stuff. DNA is much more stable than RNA so that’s a plus. The electrical mechanism for getting it in the targeted cells is unique. Looking forward to seeing the trial results.

I have a truly serious question. Is there any technology evolution or improvement you would support. Are all vaccines bad. Is there is a more elegant way to vaccinate is there a problem with it.

Science evolves. Sometimes slowly. Sometime with great lurches forward. It seems perplexing that we don’t celebrate progress

I used to support vaccinations, but am in the process of reconsidering that support.

I think of the many medical solutions/interferences over our history, such as bloodletting, thalidomide, other approved drugs whose side effects are worse than the condition for which they are prescribed, the dangers of anesthesia... and my faith in western medicine is quickly waning.

Do you ever wonder if 50 years from now we will look back on vaccinations the way we did bloodletting, and realize that we shouldn’t have been messing with our immune system, thinking we “knew” better than God’s design?

I believe in science, but it can be used for good or evil, as we can see happening today.

Smallpox was introduced into human populations about 3,500 years ago. In its last 100 years alone, it killed 500 million people. A 10 year long mass vaccination campaign wiped it off the face of the globe. The only known smallpox left on Earth is stored in laboratories for future study. No cases have been observed in humans since the 1970s. We don’t even vaccinate against smallpox anymore because it’s just gone. 3,500 years and then gone in the blink of an eye.

Polio entered the human population around 3,000 years ago. In that time, it killed a lot of children and paralyzed a lot more. As recently as 1996, polio was paralyzing over 75,000 children in Africa each year. By 2016, Africa was declared free of wild polio. Pakistan and Afghanistan are the only places left on Earth where you can find wild polio because everywhere else vaccinated it out of existence. The last case of polio acquired in the US was in 1979. It’s gone. No more iron lungs. No more paralyzed and disfigured children.

Bubonic plague wiped out 1/3 of Europe’s population. Antibiotics would have stopped it in its tracks.

Cancer mortality rates rates overall have decreased by 20% since the 1950s. Some types of cancer have seen survival rates soar with new treatments. In the 1970s, a woman had about a 70% chance of surviving 5 years after a breast cancer diagnosis. That’s over 90% today.

If you want to cherry pick a handful of mistakes in the hundreds of years of medical science advancement, you can certainly find some things to complain about. But when you look at the totality of what modern medicine has done, the net-net is hundreds of millions of deaths and lifelong debilitating injuries prevented.

You want to talk about God’s design? God gave man brains bigger than dogs so that man would rule over nature. That includes viruses, bacteria, and other pathogens. We have brains to study, observe, and plan ahead. We have hands useful for wielding tools and improving tools. We have language and culture to help pass down knowledge so incremental improvements can happen over time. That’s God’s plan; not some hippie commune where all modern human achievements are cast aside until daddy’s money runs out and the game is over. If you want to see what life looks like without the benefits of modern medicine and science, move to rural Pakistan or Afghanistan. Sure, it’ll be miserable and your life expectancy drops in half, but at least there you can yield to nature and let Jesus take the wheel.

God has a plan, and it doesn’t involve us throwing our hands up and claiming we have zero control over anything. If you’re driving down the highway at 70mph and take your hands off the wheel, your life isn’t in God’s hands (or Jesus’), but rather your soon-to-end life is in your own hands as you choose to use your God-given free will to make a foolish decision to suddenly stop steering your vehicle.

God gave us dominion over nature. Not with some snap-your-fingers magic powers, but with a brain and hands that enable us to better ourselves and control our environment in a way that no other living thing created by God can do.

Just don’t try to Godshame me or others into getting the jab.

If or when God tells me to get the jab I will.

Until then I think I’ll just wait and see

if I can trust the experts or not,

they seem to have a pretty bad batting average.

I’m absolutely not “Godshaming” anyone into getting vaccinated. If you want to get vaccinated, go right ahead. If you want to wait, wait. If you don’t want it at all, that’s fine too. I chose to get vaccinated. If you make a different choice, there’s absolutely nothing wrong with that. I 100% support the right of each person to make their own medical decisions.

I was responding to the overall anti-medicine sentiment in the original post (”we shouldn’t have been messing with our immune system, thinking we “knew” better than God’s design?”). God gave us brains and the ability to both invent and make use of tools, and the ability to improve upon those tools over generations. Tools are what give us the promised dominion over nature. The idea that we should have allowed Smallpox to kill another 500 million people because that’s what God wants is absurd. So is the notion that we should have allowed polio to continue paralyzing millions of children. I don’t know whose “god” enjoys seeing paralyzed children, but the one I worship isn’t a monster.

If we have the tools to save lives and improve lives, those tools should be made available to all who want them. If someone doesn’t want them, that’s on them. Same as if you offered someone a ride when they said they’re going somewhere 200 miles away and they refuse. You want to walk? Walk.
