STUDY: The Binding mechanism of Ivermectin and levosalbutamol with spike protein of SARS-CoV-2

ABSTRACT:

In this study, we have investigated the binding mechanism of two FDA approved drugs (ivermectin and levosalbutamol) with the spike protein of SARs-CoV-2 using three different computational modeling techniques. Molecular docking results predict that ivermectin shows a large binding affinity for spike protein (-9.0 kcal/mol) compared to levosalbutamol (-4.1 kcal/mol).

Ivermectin binds with GLN492, GLN493, GLY496 and TRY505 residues in the spike protein through hydrogen bonds and levosalbutamol binds with TYR453 and TYR505 residues. Using density functional theory (DFT) studies, we have calculated the binding energies between ivermectin and levosalbutamol with residues in spike protein which favor their binding are βˆ’ 17.8 kcal/mol and βˆ’ 20.08 kcal/mol, respectively.

The natural bond orbital (NBO) charge analysis has been performed to estimate the amount of charge transfer that occurred by two drugs during interaction with residues. Molecular dynamics (MD) study confirms the stability of spike protein bound with ivermectin through RMSD and RMSF analyses.

Three different computer modeling techniques reveal that ivermectin is more stable than levosalbutamol in the active site of spike protein where hACE2 binds.

Therefore, ivermectin can be a suitable inhibitor for SARS-CoV-2 to enter into the human cell through hACE2.

Saw a graph, today, that showed the use of IVM in India. Deaths declined. Then when they started vaccinating...the deaths went through the roof.

Personally I think the 75% reduction in mortality is largely due to late administration. I suggest that if it was given earlier the numbers would be even better. Hard to know of course since there is a time lapse between infection, symptoms, and testing. But mostly because many of these studies used severely ill patients. Gotta see how the meds work when given early. And since it would be inhumane and unethical to deny these meds even that data could only be compared to historical rates of hospitalization and death.

I always thought they should’ve been handing out a “tear open in case of emergency” bags of pills at the testing center. If you got a positive result, tear open and call the doctor for more.

Of course, if Ivermectin were shown to work in a large trial, then there would have been no money thrown at vaccines and the FDA would not have given authorization to any of them. So the powers that be intentionally spiked as much data about HCQ+ and I+ that they could reasonably cheat, using crapily designed trials and other forms of data manipulation to influence ‘science’ and perpetuate falsehoods and half-truths throughout the media.

The nurse practitioner in our doctor’s office gave it to my wife and I when we had Covid in March. I think it did a lot of good.

So they believe (and apparently have demonstrated) that ivermectin acts as a 3CL Protease Inhibitor. There are a few other drugs that can do this, as well as a couple more being trialed. I read a study a while back that determined that while coronaviruses were able to mutate and reproduce around 3CL inhibitors, that the resulting mutations were significantly weaker and reproduction reduced.

Researchers have known for some time that 3CL PI’s are a potential treatment against coronaviruses. In fact, Pfizer’s 3CL PI currently in trials is based on a molecule they were developing for SARS-1 back in 2003, but the virus disappeared and the project was shelved. I’m not yet fully convinced on the efficacy of ivermectin vs COVID in the real world, but it looks hopeful.

Great Find. If that's true, then I think Ivermectin would likely interfere with the vaccines developing a response, if you are taking it when you are vaccinated.

But as a first line of defense it should be great.
