GlyNAC improves multiple defects in aging to boost strength and cognition in older humans (Some defects “reversed to the levels found in young adults” - eGFR up 15%, IL-6 down 77%, TNFa down 40%, Fasting Insulin down 55%, etc.)

A pilot human clinical trial conducted by researchers at Baylor College of Medicine reveals that supplementation with GlyNAC—a combination of glycine and N-acetylcysteine as precursors of the natural antioxidant glutathione—could improve many age-associated defects in older humans.

The results of this study show that older humans taking GlyNAC for 24 weeks saw improvements in many characteristic defects of aging, including glutathione deficiency, oxidative stress, mitochondrial dysfunction, inflammation, insulin resistance, endothelial dysfunction, body fat, genomic toxicity, muscle strength, gait speed, exercise capacity and cognitive function. The benefits declined after stopping supplementation for 12 weeks. GlyNAC supplementation was well tolerated during the study period.

Animal studies conducted in the Sekhar lab have shown that restoring glutathione levels by providing GlyNAC reverses glutathione deficiency, reduces oxidative stress and fully restores mitochondrial function in aged mice.

"We worked with eight older adults 70 to 80 years of age, comparing them with gender-matched younger adults between 21 and 30 years old," Sekhar said. "We measured glutathione in red-blood cells, mitochondrial fuel-oxidation, plasma biomarkers of oxidative stress and oxidant damage, inflammation, endothelial function, glucose and insulin, gait-speed, muscle strength, exercise capacity, cognitive tests, gene-damage, glucose-production and muscle-protein breakdown rates and body composition. Before taking GlyNAC, all these measurements were abnormal in older adults when compared with those in younger people."

The older participants took GlyNAC for 24 weeks, and then stopped it for 12 weeks.

"We are very excited by the results," Sekhar said. "After taking GlyNAC for 24 weeks, all these defects in older adults improved and some reversed to the levels found in young adults." The researchers also determined that older adults tolerated GlyNAC well for 24 weeks. The benefits, however, declined after stopping GlyNAC supplementation for 12 weeks.

easily available via NAC precursor, but pure oral glutathione will be digested in the gut ...

Can TMG be used instead of “plain” glycine? I already take a teaspoon in my coffee every morning, and 1200 mg of NAC every other day.

I don’t know, but the NAC needs to be taken with the glycine source, and the amount of glycine needs to be equivalent to the amount of NAC.

You still can get NAC as well as glycine. I buy them both in powder form. I have been taking them along with several other nutrients for a long time. Usually about 1000 mg of NAC along with 2000 mg of ascorbic acid as for NAC to do its job it should always be taken with at least two to three times as much ascorbic acid (take buffered C) along with about 2000mg of glycine, which amongst other things if you have enough in your system prevents you from getting a sunburn. I have been dealing with them for many years, they ship prompt, have good products and are honest and are reasonably priced. Following are some characteristics of NAC

N-Acetyl Cysteine

N-Acetyl Cysteine (NAC) is a metabolite of the sulfur-containing amino acid, Cysteine. Cysteine is found in high protein foods, N-Acetyl Cysteine is not. N-Acetyl Cysteine is produced within the human body. Cysteine plays a role in the sulfation cycle, acting as a sulfur donor in phase II detoxification and as a methyl donor in the conversion of homocysteine to methionine. Cysteine also helps synthesize glutathione, one of the body’s most important natural antioxidants and detoxifiers. N-Acetyl-Cysteine is the acetylated form of L-Cysteine.

N-Acetyl Cysteine increases Glutathione Levels
N-Acetyl Cysteine is rapidly metabolized to intracellular glutathione. Glutathione acts as a powerful antioxidant in the body. Glutathione also detoxifies chemicals into less harmful compounds. N-Acetyl Cysteine also protects the body from acetaminophen toxicity and is used in hospitals for patients with acetaminophen poisoning. It has also been shown to be effective at treating liver failure from other causes as well.

N-Acetyl Cysteine Chelates Heavy Metals
Heavy metals like lead, mercury and arsenic are detoxified and removed from the body by N-Acetyl Cysteine . It also increases the excretion of zinc and other essential minerals when taken over an extended period. It is therefore necessary to supplement zinc, copper and other trace minerals when taking N-Acetyl Cysteine.

N-Acetyl Cysteine and the Immune System
Glutathione is known to aid in the transport of nutrients to lymphocytes and phagocytes, two major classes of immune cells, and to protect cell membranes. While purified glutathione is available as a dietary supplement, absorption is low, and N-Acetyl Cysteine is thought to be a better method of boosting cellular glutathione levels. Double-blind research has found that N-Acetyl Cysteine supplements improved symptoms and prevented recurrences in people with chronic bronchitis. N-Acetyl Cysteine at a dosage of 1,200 mg per day helps to prevent Influenza infection, reduces the symptoms of existing Influenza infection and reduces the duration of Influenza infections.

N-Acetyl Cysteine and Cancer
N-Acetyl CysteineN-Acetyl Cysteine has been shown to reduce the proliferation of certain cells lining the colon and may reduce the risk of colon cancer in people with recurrent polyps in the colon. Its action as an antioxidant and a glutathione precursor may also contribute to a protective effect against cancer.

N-Acetyl Cysteine Cautions
When taking N-acetyl cysteine it is recommended that two to three times as much vitamin C be taken at the same time. Failure to do so may result in more harm than good from taking this product because of the prolonged presence of the oxidized form of L-Cysteine. The vitamin C also helps keep the glutathione that is produced from the Cysteine in its reduced form so that it can continue acting as an antioxidant.

N-Acetyl Cysteine Dosage
Typical dosage recommendations are in the range of 250-1500mg of NAC daily for the majority of therapeutic benefits.

N-Acetyl Cysteine Safety
NAC is considered safe for consumption in its therapeutic dosage ranges. Individual reactions may vary and anyone experiencing adverse symptoms while taking N-Acetyl Cysteine should discontinue it.

Some they say works best by themselves on an empty stomach with water.

Some do better taken with a meal.

I would suspect the empty stomach w/water ones are water soluble, and the ones taken with a meal are fat soluble.

In lifespan studies in mice, the 24% improvement was the exact same for both GlyNAC and N-acetylcysteine itself. And as mentioned above it’s much cheaper just to buy the amino acids themselves rather than this overpriced hyped pill.

Glutathione is a 3-amino-acid-long chain. I’ve read it gets broken up in the digestive tract.

I’ve also read that whey protein powder stimulates the body to make glutathione at the cellular level.

YMMV.

It doesn’t survive digestion. You’d have to inject it. I’ve done both. NAC works just as well.
