Subject: Out of town pool report #2 — Phoenix arrival

Air Force One touched down at Phoenix Sky Harbor International Airport at 12:04 p.m local time. The flight was relatively uneventful. Chief of Staff Mark Meadows popped back into the press cabin to say a brief hello during the flight.

POTUS deplaned at 12:20 p.m. He was greeted by Arizona Gov. Doug Ducey.

It is hot and sunny in Phoenix. We are rolling to Honeywell International Inc. in Phoenix.

As many of you may have noticed, VP Mike Pence and coronavirus task force members conducted a briefing for reporters this afternoon.

The pen and pad was not on his public schedule and pooler was not among those invited to attend. It was held at OEOB and included wires, some conservative-leaning outlets, and other media. Public reporting indicates Pence confirmed discussions of disbanding the task force. A transcript is expected to be forthcoming, according to Pence’s office. Pooler will seek to share when it is out.

Also, Brian Kerem sends along a link to a brief gaggle that Kevin Hassett gave on the north lawn that was simultaneous with the POTUS departure.

Chloroquine Is a Potent Inhibitor of SARS Coronavirus Infection and Spread

Abstract
Background: Severe acute respiratory syndrome (SARS) is caused by a newly discovered coronavirus (SARS-CoV). No effective prophylactic or post-exposure therapy is currently available.

Results: We report, however, that chloroquine has strong antiviral effects on SARS-CoV infection of primate cells. These inhibitory effects are observed when the cells are treated with the drug either before or after exposure to the virus, suggesting both prophylactic and therapeutic advantage. In addition to the well-known functions of chloroquine such as elevations of endosomal pH, the drug appears to interfere with terminal glycosylation of the cellular receptor, angiotensin-converting enzyme 2. This may negatively influence the virus-receptor binding and abrogate the infection, with further ramifications by the elevation of vesicular pH, resulting in the inhibition of infection and spread of SARS CoV at clinically admissible concentrations.

Conclusion: Chloroquine is effective in preventing the spread of SARS CoV in cell culture. Favorable inhibition of virus spread was observed when the cells were either treated with chloroquine prior to or after SARS CoV infection. In addition, the indirect immunofluorescence assay described herein represents a simple and rapid method for screening SARS-CoV antiviral compounds.

Methods
We did next-generation sequencing of samples from bronchoalveolar lavage fluid and cultured isolates from nine inpatients, eight of whom had visited the Huanan seafood market in Wuhan. Complete and partial 2019-nCoV genome sequences were obtained from these individuals. Viral contigs were connected using Sanger sequencing to obtain the full-length genomes, with the terminal regions determined by rapid amplification of cDNA ends. Phylogenetic analysis of these 2019-nCoV genomes and those of other coronaviruses was used to determine the evolutionary history of the virus and help infer its likely origin. Homology modelling was done to explore the likely receptor-binding properties of the virus.
Findings
The ten genome sequences of 2019-nCoV obtained from the nine patients were extremely similar, exhibiting more than 99·98% sequence identity. Notably, 2019-nCoV was closely related (with 88% identity) to two bat-derived severe acute respiratory syndrome (SARS)-like coronaviruses, bat-SL-CoVZC45 and bat-SL-CoVZXC21, collected in 2018 in Zhoushan, eastern China, but were more distant from SARS-CoV (about 79%) and MERS-CoV (about 50%). Phylogenetic analysis revealed that 2019-nCoV fell within the subgenus Sarbecovirus of the genus Betacoronavirus, with a relatively long branch length to its closest relatives bat-SL-CoVZC45 and bat-SL-CoVZXC21, and was genetically distinct from SARS-CoV. Notably, homology modelling revealed that 2019-nCoV had a similar receptor-binding domain structure to that of SARS-CoV, despite amino acid variation at some key residues.

When the protein sequence of the SARS-CoV-2 receptor binding site was analyzed, an interesting result was found. While SARS-CoV-2 is overall more similar to bat coronaviruses, the receptor binding site was more similar to SARS-CoV.

Summary
The emergence of SARS-CoV-2 has resulted in >90,000 infections and >3,000 deaths. Coronavirus spike (S) glycoproteins promote entry into cells and are the main target of antibodies. We show that SARS-CoV-2 S uses ACE2 to enter cells and that the receptor-binding domains of SARS-CoV-2 S and SARS-CoV S bind with similar affinities to human ACE2, correlating with the efficient spread of SARS-CoV-2 among humans. We found that the SARS-CoV-2 S glycoprotein harbors a furin cleavage site at the boundary between the S1/S2 subunits, which is processed during biogenesis and sets this virus apart from SARS-CoV and SARS-related CoVs. We determined cryo-EM structures of the SARS-CoV-2 S ectodomain trimer, providing a blueprint for the design of vaccines and inhibitors of viral entry. Finally, we demonstrate that SARS-CoV S murine polyclonal antibodies potently inhibited SARS-CoV-2 S mediated entry into cells, indicating that cross-neutralizing antibodies targeting conserved S epitopes can be elicited upon vaccination.

Both SARS-CoV-2 and SARS-CoV use the same host cell receptor. It also found that, for both viruses, the viral proteins used for host cell entry bind to the receptor with the same tightness (affinity).

No. I got some updated info from my niece, who’s boyfriend works at the Honeywell engine campus, where a news report said P. Trump was going to tour. The bf said POTUS was not coming to the engine campus but going to another bldg. which was converted to making the masks. I had reconnoitered the engine campus and neighborhood for plans to position myself in the general vicinity where I could find parking and walk to the roadway that POTUS would use to reach the campus.

With last minute information that the bldg. to be toured was a different location, nearer to Honeywell’s HQ’s (I think) I decided to pass on the event. I anticipated that the police would block off the streets, some distance from the first location, and I planned my parking strategy accordingly. With a shift in the location, and time slipping by, I talked myself out of going due to a completely different lay of the land.

I was outside at 11:30 watching overhead for AF1. Once 11:40 passed, I went back inside thinking that AF1 had landed. Turns out, AF1 was still inbound at that time and landed at 12:04.

Looks like the news coverage will be plentiful. Watching FOX 10.

Getting an update on the bldg. location. It’s a structure directly on the airport property. I would have definitely been in the wrong location but only off physically by about two blocks, divided by fencing and obvious airport trespassing restrictions.

CNN sends Trump campaign cease-and-desist letter for misleading ad

The Trump ad, titled “American Comeback,” was introduced on Sunday night. The campaign says it is spending “mid-seven figures,” at least several million dollars, to run the ad on cable and broadcast channels.

The ad takes a segment from CNN’s “The Situation Room” out of context. On March 30, anchor Wolf Blitzer asked Dr. Sanjay Gupta, “Well, is it accurate that if these steps had not been put in place, the stay at home orders, the social distancing orders, as the President said yesterday, it could have been 2 million people down here in the United States?”

Blitzer’s question was about “stay at home orders” and social distancing, but in the campaign ad, that portion of his question was omitted. After Blitzer and Gupta are shown on screen, the ad cuts to taped video that invokes Trump’s restrictions on travel from China, like an airport screen showing cancelled flights.

Blitzer is heard asking, “Is it accurate that if these steps had not been put in place ... it could’ve been 2 million people dead here in the United States?” And Gupta is heard saying, “yes.”

In a statement to CNN Business, Murtaugh responded: “No discussion of efforts to prevent American deaths from the coronavirus can be had without the understanding that President Trump restricted travel from China in January. Based on that alone, the ad is accurate.”

In Murtaugh’s response on Monday evening, he said CNN is “once again the only outlet to reject a Trump campaign ad, and has now rejected multiple Trump ads that are demonstrably accurate.” He called the network “effectively a Joe Biden Super PAC,” yet another signal that anti-media rhetoric is a major part of Trump’s re-election strategy

After a very short drive, we arrived at the Honeywell facility at 12:26 p.m. The president is first scheduled to participate in a roundtable discussion on supporting Native Americans. Then he will tour the facility and later he will deliver remarks.

Today our Dem governor in DE announced some easing of restrictions to allow more businesses to re-open here in DE.

Not great, but some progress at least.

Our governor is getting a lot of pushback from the citizens of DE.

In this Google maps view, the Honeywell complex consisting of 4 bldgs. can be seen on Sky Harbor Airport property, just south of the intersection of S. 32nd St., and Air Lane.

My intended location was to have been at S. 34th St., and E. Jackson St.
