New study shows vitamin C boosts DNA damage and cell death in melanoma cells (With Hydrogen Peroxide)

A recent study shows that there may be a more effective way to treat melanoma by using ascorbate (vitamin C) to increase DNA damage in cancer cells, leading to their death, according to Marcus Cooke.

The multi-disciplinary research team found that melanoma cells have more DNA damage and fewer antioxidant defenses compared to normal skin cells. When treated with hydrogen peroxide and vitamin C, melanoma cells showed even more DNA damage and higher cell death rates, while normal cells were protected. Additionally, the study results found that vitamin C boosted the effectiveness of an existing melanoma drug, elesclomol.

"We've been studying the effects of antioxidants since the late 1990s and have been fascinated by the ability of vitamin C to act as a prooxidant (causing DNA damage) and antioxidant (preventing DNA damage), and its apparent ability to help modulate DNA repair," Cooke said.

"The findings show that melanoma cells have higher levels of DNA damage than keratinocytes (the primary type of cell found in the epidermis). We discovered that this damage was proportional to the amount of melanin that is in the melanocytes—the more melanin, the more damage," he explained. "This is in cells not even exposed to sunlight, suggesting that melanin within cells can cause damage in melanoma cells."

Cooke acknowledges that additional clinical studies and trials would bolster these findings and assist in moving towards including ascorbate as an accessory in treatment.

"Given that ascorbate is already well studied and is known to be well tolerated, I think that clinicians could include ascorbate as an addition to existing treatment to enhance existing approaches if they act by inducing DNA damage as elesclomol does," he said.

I understand that vitamin C can simply be taken by mouth, but how is the hydrogen peroxide administered to in-vitro melanoma?

Topically?

I ask, because I’ve had two melanomas removed by surgery.

Don’t melanomas sometimes heal themselves and disappear?

Pauling and Cameron in the 1970s showed that intravenous C-vitamin increased survival rates in patients with terminal cancer. Their study was not placebo controlled, so when the Mayo Clinic published two placebo controlled studies that neither showed any effect the interest in use of C-vitamin in cancer treatment faded. The fact that the latter studies used only oral administration and for shorter periods was considered less important than the fact the studies originated from a very reputable institution and were placebo controlled.

I'm going to take this with a huge grain of salt. This is because the study used cells in culture. All kinds of things kill cancer cells in culture but do not translate to any kind of benefit in a living patient. I used to use bleach to kill cancer cells in culture; there is no way to bleach a patient to kill the cancer without killing the patient.

The multi-disciplinary research team found that melanoma cells have more DNA damage and fewer antioxidant defenses compared to normal skin cells.

The reason that radiation therapy is effective with cancer is that cancerous cells are less capable of repairing DNA damage than normal cells. The radiation induces extensive DNA damage. Cancer cells die and normal cells repair themselves (or decide to die also).

When treated with hydrogen peroxide and vitamin C, melanoma cells showed even more DNA damage and higher cell death rates, while normal cells were protected. Additionally, the study results found that vitamin C boosted the effectiveness of an existing melanoma drug, elesclomol.

Normal cells are not resistant to hydrogen peroxide (H2O2). It is difficult to imagine how H2O2 could be administered to an intact patient without having all kinds of toxicity issues. It might be beneficial to make sure patients get adequate vitamin C during chemotherapy--but trying to load up their bodies with excess vitamin C is likely to be a futile approach, since their kidneys will be working to remove all excess vitamin C.

A few citations on this thread by NIH. During Covid, we learned that that organization has corrupt leadership, and is in bed financially with drug companies. Since then, it’s hard to put much faith in anything they say or do.
