The scene at the end of "Wait Until Dark" where Alan Arkin and Audrey Hepburn struggle in total darkness. You cannot see what is happening which adds exponentially to the tension.

Though this choice may not be too well known, my second choice goes to John Goodman and Jeff Bridges in The Big Lebowshki. The "Mark it zero" scene in the bowling alley is just GREAT!!

Shane ... one of the best westerns ever ... Brandon De Wilde (a young boy) follows Shane into town from his parents homestead, calling out after him 'Shane ... Shane ... come back'. Shane rides on into town ... kills the bad guys (one of them a young Jack Palance) ... and rides off into the evening darkness.
This movie was filmed near Jackson Hole, WY in 1953 ... the scenery with the Tetons in the background is magnificent.
