Return of the Pink Panther:
Clouseau: "Does your dog bite?"
Man: "No."
Clouseau: "Nice doggy!"
Dog bites Clouseau and barks
Clouseau: "I thought you said your dog did not bite!"
Man: "That is not my dog."

And for some reason, I keep remembering the following from the forgetable movie Best Friends:

Col. Nathan Jessup: Son, we live in a world that has walls, and those walls have to be guarded by men with guns. Who's gonna do it? You? You, Lieutenant Weinberg? I have a greater responsibility than you can possibly fathom. You weep for Santiago and you curse the marines. You have that luxury. You have the luxury of not knowing what I know: that Santiago's death, while tragic, probably saved lives. And my existence, while grotesque and incomprehensible to you, saves lives. You don't want the truth because, deep down in places you don't talk about at parties, you want me on that wall, you need me on that wall. We use words like honor, code, loyalty. We use these words as the backbone of a life spent defending something. You use them as a punchline. I have neither the time nor the inclination to explain myself to a man who rises and sleeps under the blanket of the very freedom that I provide and then questions the manner in which I provide it. I would rather you just said "thank you" and went on your way. Otherwise I suggest you pick up a weapon and stand at post. Either way, I don't give a damn what you think you are entitled to.

My favorite is not a line from the script, but a line from the lyrics "High Noon" for the movie of the same name:
...I do not know what fate awaits me
I only know I must be brave
And I must face the man who hates me
Or lie a coward, a craven coward,
or lie a coward in my grave...

"I want my father back, you son of a bitch." [as Inigo stabs Count Rugen] - Princess Bride
The best, hate-filled, kill-the-bad-guy line in all of film, perfectly delivered by Mandy Patinkin.

My favorite line is from the first Muppet Movie. Steve Martin says,
"Oh, may I?" It comes in handy with my sixth graders.
Favorite gesture (no, not what you may think!) in a movie comes from The Sting: laying a finger aside of the nose indicating that all the plans are in place and working.

I can't remember the film, but:
Kid: You shot him in the back!
Gunslinger: His back was to me.

"How'd you like to take a ride on the wild baloney pony?"
"Would you like some of my man-sized manicotti?"
"I've been to the Hawaiin islands, ever heard of Comeoniwannalayya?"
