I run both MS Anti-Spyware and SpyBot Search & Destroy.
Each has a couple of features that the other does not as is usually the case with software.
For example, MS Anti-Spyware updates its database more often and prevents more spyware sites. Spybot's database is not updated as often.
However, SpyBot Search & Destroy also monitors attempted, unauthorized changes to critical system files. MS Anti-Spyware does not have this feature.
I see nothing wrong with running both versions of Anti-Spyware as long as you have sufficient system resources, ( Physical RAM memory on the motherboard. )
If you have some other version of anti-spyware, I say, "Run both because in all likelihood one has features that the other doesn't and vicey versy." :-)

Forgot to mention that MS Anti-Spyware updates automatically if you are connected to the internet. SpyBot has to be updated manually.
