An oral supplement intended to stimulate a natural body process appears to promote muscle endurance and mitochondrial health in humans. New research suggests that the supplement, urolithin A, may help improve or prolong muscle activity in people who are aging or who have diseases that make exercise difficult.
Urolithin A is a byproduct of a person's gut bacteria and a diet comprising polyphenols found in pomegranates, berries and nuts. The compound also is produced and sold by dietary supplement companies.
Supplemental urolithin A has been shown in animal tests and molecular studies of humans to stimulate mitophagy, a process that Marcinek explained as "mitochondrial quality control."
"Mitochondria are like batteries that power the cells in your body," he said. "But over time they break down. The process of mitophagy recognizes this failure and proactively tears down the mitochondria, reducing it to elemental components that a cell can reuse. But with aging, mitophagy becomes less efficient and your body accumulates this pool of failing mitochondria. It's one way that muscles become less functional as we age."
The researchers studied a small cohort of people over age 65 who were randomized to receive a placebo or a daily supplement of 1,000 mg urolithin A for four months.
Measures of distance covered during a six-minute walk improved markedly between tests at baseline and four months in both the supplement and placebo group.
He added that urolithin A supplements could have potential to benefit people who cannot get the exercise they want due to poor muscle health or disease.
"Just getting them over that point where exercise is possible—a walk around the block or climbing some stairs—might help a person build their own health."

Both groups improved over 4 months because they were walking everyday.

There actually was a very sizable change with endurance.
Am I understanding correctly that the placebo group produced similar improved results.after 4 months?

Muscle endurance (ie, resistance to fatigue) was assessed on the right hand (FDI) and leg skeletal (TA) muscles by measuring the number of contractions during repeated isometric contractions until fatigue using a custom-built exercise apparatus19,20 at baseline, 2 months, and 4 months. The contraction intensity was set at 70% of the maximum voluntary contraction, and the exercise began at a frequency of 60 contractions per minute (cpm) for the first minute and increased at a rate of 10 cpm each minute until exhaustion.
To assess the effect of urolithin A supplementation on muscle endurance, we measured the ability to maintain repeated voluntary contractions to 70% of maximum voluntary contraction in the FDI and TA. At the 2-month visit, urolithin A supplementation significantly improved endurance in both muscles compared with placebo (urolithin A: FDI, 95.3 [115.5] and TA, 41.4 [65.5]; placebo: FDI, 11.6 [147.4] and TA, 5.7 [127.1]) (Figure 2D-I; eFigure 2A and B in Supplement 2). At the 4-month visit, endurance in the urolithin A group continued to improve, but a parallel increase in the placebo group led to no significant effect of treatment.
We further analyzed plasma samples to investigate the effect of urolithin A on biomarkers of mitochondrial health and inflammation. Urolithin A supplementation led to a significant reduction in several acylcarnitines, primarily long-chain and polyunsaturated species, at both the 2-month and 4-month visits compared with baseline (Figure 3A-D; eFigure 4A and B in Supplement 2). In addition, several species from a panel of ceramides that were analyzed via plasma metabolomics showed a significant reduction from baseline measurement in the urolithin A group but not the placebo group at both 2 and 4 months (Figure 3E and F; and eFigure 4C and D in Supplement 2).
To further test the effect of urolithin A on inflammation, we measured plasma levels of the inflammatory biomarker CRP. As for ceramides, a significant reduction from baseline in plasma CRP was observed in participants in the urolithin A group but not the placebo group (baseline vs 4 months: urolithin A, 2.14 [2.15] vs 2.07 [1.46]; placebo, 2.17 [2.52] vs 2.65 [1.86]) (Figure 3G).
If people took every supplement advertised they’d have to swallow about a thousand pounds of crap a day.
Agreed, but targeting them for your specific circumstances is what counts.

Yup me too. Desperate people buy these things thinking it’s gonna make them well again, but nope it has very little if any effect many times. The results don’t match the claims. No saying this,product isn’t good, but I just don’t know. I have taken ,ots of stuff based on reviews, wasted lots of money, and not getting the results claimed by the sellers.
I'd rather spend $50 every so often on pomegranites, berries and nuts and eat them regularly. Aldi has reasonable-cost options for all kinds of berries and nuts, and pomegranites in the Thanksgiving thru Christmas season (and occasionally bottled pomegranite juice).
