A number of studies have linked TMAO to heart disease, however, until now it hasnt been clear exactly how this metabolite causes cardiovascular damage. A robust new study, published in the journal Hypertension, is offering one of the first thorough mechanistic investigations illustrating how TMAO damages the cardiovascular system.
Our work shows for the first time that not only is this compound directly impairing artery function, it may also help explain the damage to the cardiovascular system that naturally occurs with age, says Vienna Brunt, first author on the new study.

Some time back there was an article that said studies based on mice & rats were found NOT to apply very well to humans. Now if this article said that post-mortem studies had found increased levels of TMAO in humans who died of heart disease, I might take it more seriously.

The new research examined data from a large longitudinal study known as the Nurses Health Study, which followed over 100,000 nurses for a decade. This data offers some of the first insights into the long-term relationship between TMAO blood levels and cardiovascular health. Over 10 years a distinct link between increased TMAO levels and coronary heart disease (CHD) was detected. Women with the largest TMAO blood level increase across the 10-year period displayed a 63-percent higher risk of CHD.

I recall reading recently about a large-scale study of the drug Metformin - it showed that diabetics given this lived years longer than those without the disease! It can also reduce inflamation, improving outcomes for cancer and heart treaments.
