Most people became familiar with the phrase from the movie Tombstone, as it's uttered by Val Kilmer as Doc Holiday.

How we got the phrase is a long story, but according to World Wide Words:

What it means is easy enough. To be one’s huckleberry — usually as the phrase I’m your huckleberry — is to be just the right person for a given job, or a willing executor of some commission.
