Isadora said that a man who identifies as a woman is still “born male” and “retains their birth DNA.”

No a man who is born a man with an "X" chromosome retains HIS birth DNA, even if he chops off a body part.

All men get an X chromosome, from their mother. He is male because he received the Y chromosome from his dad. Chopping off or otherwise modifying body parts doesn’t change his DNA. If it still exists hundreds of years from now, his skeleton will still be a male skeleton.
