No a man who is born a man with an “X” chromosome retains HIS birth DNA
All men get an X chromosome, from their mother. He is male because he received the Y chromosome from his dad. Chopping off or otherwise modifying body parts doesn’t change his DNA. If it still exists hundreds of years from now, his skeleton will still be a male skeleton.

Right, because HE normally only has one x chromosome. Some have two X chromosomes and a Y chromosome, but then HE is still a male.
