I was wondering, did the American Colonist register weapons before Lexington and Concord? How did the British know where the cache of weapons and gun powder were at?

Not registered. Ordered confiscated by the military governor so that the inhabitants of the rebellious city of Boston could not raise their hands in defiance to His Sovereign Majesty, George III, King of Great Britain, Scotland, Wales and Ireland.

There were a couple of British officers assigned to locate caches out in the countryside, also a couple of loyalists aiding them. I think one of the loyalists was a Concord resident, which is why they knew almost exactly where at least a portion of that particular stockpile was. The British officers on that duty were the same gents who captured Paul Revere, I believe.

On an unrelated but interesting note, the minute men (not the regular militia though) were under salary by the crown at the time of the battle.
