The Tory leader travelled to Cardiff to celebrate the results after his party won the largest share of the Welsh popular vote in the European election.
The disastrous result cost Labour an MEP and leaves it, the Conservatives, Plaid Cymru and the UK Independence Party with one Welsh seat each in the European parliament.
Labour blamed its dire showing on voters staying at home because of a "toxic combination" of anger over MPs' expenses and the recession. Mr Cameron said: "Since the time of Lloyd George has a party other than Labour won in Wales and today it is the Conservative Party.
"No longer should people say in politics that there are uncrackable bastions, safe seats where Conservatives cannot go.
"There will be a great toppling of Labour MPs if these results were copied at a general election."
Labour won 20.3 per cent of the vote in Wales, down 12.2 percentage points on the last European election in 2004.
The Tories were on 21.2 per cent, up 1.8 percentage points, while Plaid Cymru improved its performance by 1.1 percentage points to 19 per cent.

He said: "We've had an even bigger one today. It's off the Richter scale. The political landscape has changed here."

Next president - Sarah Palin.
Ping.
Wonderful news. I agree, this means Palin will be elected president in 2012, and America and the World will be better for it. Who should she pick as VP? I hope she can pull in a CONSERVATIVE Republican congress too.
Just a few more years till sanity returns. We can hold out that long.
Snubbing traditional allies might have consequences here.
The way things are going I’m not sure we can last to the end of 2009...sanity-wise.
No, no, no!!! Don’t take the wrong message. The voters were angry because you didn’t kiss the 0ne’s arse enough. Just keep it up, only more so, that’s the ticket, lads and lassies! If you grovel at his feet you’ll, well, let’s say we’ll be amazed at the results of the next elections.
In other wonderful news, the Newspaper Guild rejected the New York Times ultimatum to keep the Boston Globe open. Mother of God! Is this the end of Rico?
Hear hear!
Conservatism is over? Uh, I don't think so. (And if these results make Pelosi & Crowd pinch just a bit harder, so be it...)
This encourages me further to Battle On!
Wasn't he in Paris long enough for EVERYONE to participate? How about Cairo? Saudi Arabia?
That is one worn out behind.

WOW! Would that be the new 'old' plague - or the old 'new' plague?
Labour was given a fierce pimp slap. May the Dems receive the same in 2010 and 2012.
Pimp - bitch. This is no time to dicker about terms.
