The fastest mass extinction outside of a major meteor striking the Earth (the big ones are easy to find) is the possibility of a supervolcano eruption with volanic ash and sulfur particle output 15 to 20 times that of Mount Tambora's eruption in 1815. And more than just the Yelllowstone Caldera is capable of such an eruption--that that could completely turn the sky dark for at minimum 4-5 months effectively worldwide.
