How do you tell your employer NO when they are telling employees they need to get the shots when they work from home on computer and have NO face to face contact with employees or customers?

Depends on if you are willing to risk your job. I’m lucky in that I work only for myself from home as I’m retired and just an investor now. It’s much harder for employed folks and I sympathize with them.

My position is everyone should make their own medical choices and leave each other alone whether to get the shots or not.

Many employers do totally insane stuff.

That is your clue to polish the resume and get it out there...

Depends on your state. Here in CA and other At-will employment states, your option is to tell them know by quitting.
