The growth of humankind had to start with a lot of in-breeding. Small isolated communities grew into bigger and bigger ones without outside interaction. I grab a few paragraphs from some sites for reference. But in-breeding has been going on for thousands of years.

At least 16 million men in central Asia have the same male Y chromosome as the great Mongol leader.

‘One thirteenth century Persian historian claimed that within a century of Khan’s birth, his enthusiastic mating habits had created a lineage of more than 20,000 individuals,’ said team leader Dr Chris Tyler-Smith. ‘That now appears to account for around 8% of the men in central Asia.’

The ancient Egyptian royal families (for 3000 years) were almost expected to marry within the family, as inbreeding was present in virtually every dynasty. Pharaohs were not only wed to their brothers and sisters, but there were also “double-niece” marriages, where a man married a girl whose parents were his own brother and sister. It is believed that the pharaohs did this because of the ancient belief that the god Osiris married his sister Isis to keep their bloodline pure.
