I think John Wayne used the quote on a TV variety show, not in a movie.
I could have sworn that Sgt. Stryker said it in "Sands of Iwo Jima," (I can even picture the scene) but, upon further review, it appears that you are correct and John Wayne never spoke those words in a movie.
