Calling something a "daisy" then was the equivalent of "cool", "nifty", "peachy", "the cat's pajamas" or any other such goofy, nonsensical thing we say today. So when Doc said "you're a daisy if you do", it might be like the Church Lady saying "well, isn't that special?".

Doc always assumed he would be killed in a gunfight. When the drop was on him, he was signaling that he wasn't scared to die.("You're a daisy if you do") His last words were along the lines of "Isn't this funny?" because he died with his boots off in a bed.

Oddly, Wyatt lived in LA until his death, forever dodging rumors that his character was greatly embellished.
