Well the Inquisition more than a bit like the Salem Witch trials.
In Salem, however it started, it basically became a scam to grab land from people and mess with inheritance.
The Inquisition was no different. Lots of people were caught up in it that were faithful but someone powerful owed them money. Or they were on the wrong side of someone political. And suddenly they were Jewish.

There is no doubt that Columbus was born in the Republic of Genoa to Catholic parents, both of whom perhaps had some Jewish ancestry; how much is entirely uncertain — and meaningless as to Columbus’ own identity.
(My DNA, for example, shows largely Scandinavian origins, but my ancestry itself is nearly all English.)
Whatever Columbus’ DNA suggests, real or not, is meaningless as to his Catholic identity, as he was raised in a Catholic family, and he devotedly practiced the faith, especially in terms of his legacy as explorer.

No. Neither Jews or Muslims were "tortured to death" if they did not convert.
Granted, Spanish Catholics were not feeling kindly toward Jews or Muslims, who were close allies during the Muslim conquest and occupation of Spain (which Ferdinand and Isabella ended).
And with the expulsion of the Muslim invaders, there were social and economic benefits to being a Catholic. And so some remnant Muslims and Jews pretended to convert, essentially infiltrating and undermining the church (much like today's gay mafia in the church).
The Inquisition only targeted heretics, including Catholics, but also false converts among Muslims and Jews.
But the Inquisition claimed no jurisdiction over a Muslim or a Jew who choose to remain a Muslim or Jew.
That’s really the point. Christians don’t have a DNA profile. They can have any ancestry. Europeans act like your ancestry determines what religion you are.
It’ll be interesting to see what the DNA analysis actually says.
Other times, if a person is prominent, Jews will want to claim that person even if he only has a distant Jewish relative on his father's side. Because it's good PR.
However, now that Columbus is increasingly seen as a racist white supremacist who genocided Native Americans, the PR is not so good. So I imagine that many Jews feel conflicted as to whether they should try and claim Columbus.

Somebody taught him Hebrew and how to calculate the Jewish holidays, and even certain Jewish prayers, as they were all set out in his log book.

I think you are close. Pretty sure the Jewish rule is you are automatically Jewish if your mother is Jewish, but anyone is free to covert.
