Well, the Serbs managed to shoot down an F117 with old Russian equipment. We tried to pass off the shoot down as a one in a million shot a “magic bb, but was it? So yes, they could.
There probably is a fairly large window of opportunity to create false positives and create confusion resulting in wasted launches.
I suspect that it is part of the plan.
No one thought Japan could produce the Zero fighter or attack Pearl Harbor.

the questions should be could they and would they? Maybe they could get off a lucky shot but they know the ramifications for such a thing would be huge, under any other President. But I believe Putin surely has reserved that decision for himself of some very high ranking person under strict orders to get it right before pulling the trigger.

If they have a counter, they're not going to show their hand until there's something worth letting the Russians know they have the counter; i.e. against something much more important than targets in Syria that can be attacked by artillery or commandos.
If the US were to begin air-strikes my bet is they'd first let the US go in without whatever they've worked out for themselves which then makes any effective counter they may have worth a lot more if the US is foiled, and if the US has no problems, lets them know they don't need to share whatever they have until it becomes worth more to the US.

Wouldn’t it be cool to find out. After all the only loss would be a dead pilot and possibly a third world war. A lot of damage can be done in 100 days.
3. Robot aircraft (UCAV) could be guided to them by VHF/UHF radar, then those aircraft could target them with IR missiles not unlike the Sidewinder.
The invincible F-35 cannot fly far enough to do anything - it is so range limited it would have to be positioned in Syria to be effective if it could carry any bomb load which amounted to anything.
There are no known drones which could pull off anything bomb-wise anywhere in Syria with out being shot down within seconds of crossing the Syrian boarder.
