What I want to know is, who is the lucky bloke who gets to count these spores and what counting method do they use?

This is only a guess, but I believe they would take a small sample and count the amount of spores within it. Then using the premeasured weight of the counted spores and the premeasured weight of total spores calculate the total amount of spores in the total sample.
