No the Speaker of the House does not become vice-president.
If the office of vice-president is vacant, the president nominates a replacement, subject to confirmation by both Houses of Congress.

In the event of a vacancy, doesn't the Speaker of the House become VP?No. The VP would be nominated by the new president and would need to be confirmed by a majority of both houses of Congress. So if Biden left for whatever reason, Harris would become president. She would nominate a VP and there would be quite a show with the R House and D Senate trying to come up with a majority to confirm.
