If Manchin or Sinema were to go, their replacements would be Republicans, as the governors of Arizona and West Virginia are Republicans. That would give the GOP 52 votes, making McConnell majority leader and flipping the committee chairs to the ranking Republican. So Manchin and Sinema are not in danger.
