I believe that the Romans did that.The Eastern Gate was sealed in 1541 by the Ottoman Turks. They did it to prevent the fulfillment of the prophesy by which Messiah would enter Jerusalem. Also, the cemetery in front of the gate is a muslim cemetery built by muslims again in order to prevent Messiah from entering the City. There were no muslims when the Romans controlled Judea.
