Is it true that she said she didn’t know where her husband is? Or is that from the press?

I think she knows, but according to this article (SC governor's whereabouts unknown, even to wife (^)), she says she doesn't.

FTA:

"He was writing something and wanted some space to get away from the kids," Jenny Sanford told The Associated Press while vacationing with the couple's four sons at their Sullivans Island beach house. She said she didn't know where he was, but wasn't concerned.

Either way, it isn't being handled well, because at the end of the article, his opponents on both sides of the aisle vocally criticize him and make a point about his absence being unacceptable in terms of the National Guard, etc.

If it isn’t being handled well, it’s probably because he’s done it many times before without an issue ever arising. He’s just one Governor from 50 states. And his state isn’t NY, Texas or California so usually daily lives fly under the radar.

Cut to the chase. This gives all appearances of his political enemies on both aisles anticipating a run for the W.H. from Sanford concocting a publicity scheme to take advantage of a known absence. Blown out of proportion by everyone.

It’s a good experience for him though. If he’s really serious about a run, they’ll take things even more trivial and blow them up so he’s going to have to alter his lifestyle somewhat with an eye to that in mind.
