Hamlet's mom uses that line while watching a play Hamlet stages. The prince's intent is to guilt-trip either his mom or his uncle into confessing their adulterous murder of Hamlet's dad. Hamlet's hired actors portray his mother as initially virtuous, then succumbing to her husband's brother's lecherous advances.

After watching her thespian doppelganger's perfomance, Hamlet's mom realizes that she was way too easy, and remarks "The lady protests too much, methinks."

Thus, the quote is actually an indictment of the speaker, not (as it is usually used) of the one about whom it is spoken.
