According to the recent biopic, he was basically assaulted by a disgruntled spiritualist who ambushed him backstage where he was already ill from the inflamed appendix.
Snopes (for what it's worth) says that Houdini was likely reclining and that he received a number of brutal blows from a college-aged "fan." But it then questions whether the ruptured appendix was the result of the blunt trauma from the blows or not, noting that such ruptures are rare.
It is possible that Houdini died of a ruptured appendix (actually, from the peritonitis that resulted therefrom) that had nothing to do with his being hit in the belly.

Or he may have died from the appendicitis regardless of whether he had been struck in the abdomen or not.
But the "punch" story has that delicious irony that gives it life.

But he was lying down ill, and was not prepared when the blows, numbering at least four and forcefully delivered, fell. It wasn't like he was jumped in an alley, but the striker did not give him time to tighten his muscles in anticipation of the blows.
There is now some doubt as to whether Houdini ever even made the brag about being able to withstand any punch a man could deliver. He certainly had strong musculature and extreme flexibility, and he was no doubt a showman extraordinaire so it wouldn't be out of keeping with his persona. But that claim has never been substantiated.
