At the very least, Hillary Clinton (or more likely through her henchmen) would have raised the issue during the primary.

I believe the FBI would have done some type of Background Investigation prior to him becoming a US Senator. There has to be a file on him at the Bureau containing a National Agency Check (NAC) which would have confirmed whether or not he was a US citizen.

Since when has the Constitution been any more then a historical annoyance to the Democrat party?
If he turns out to be a non-citizen, this will be a good test to see if the Constitution has any meaning to the voters of the US. (Other then that vague remembrance of being forced to memorize the preamble back in 9th grade social studies class!)

There is no such office. Which is why the issue must be raised. Ultimately eligibility questions would go to the Supreme Court.

A candidate has to file a statement of candidacy. The govt probably accepts his word that he meets the requirements.
Have you ever applied for a professional license that asks if you have ever been in a psychiatric institution or some such thing? When you say no, the drones who process your application in most states don't check to confirm you are telling the truth. They take your word for it.
