In ancient Hebrew, Kohanim (plural) was the class of priests who presided at the Holy Temple. A Kohan was an individual priest. Nominally, Jewish people named Kohan or Cohen would be decendants of these people.

As I recall DNA studies have affirmed this and affirmed that said bloodline is more pure than other Jewish bloodlines.
