In the case of heavy trucks, as for ships, electric lines, or trains, it is the commerce clause. The reasoning, as I would put it, is thus: The constitution reserves for the federal government the right to regulate interstate commerce, this has been interpreted to include the right to regulate the conveyances that are of the type that are used for said commerce. I believe that the statute defines this for trucks as vehicles in excess of 10,000 lbs GCVW. You will find that most if not all city buses have DOT number on them, this is despite the fact that for many cities these buses cross state lines once in their lifetime (when being delivered).
