Technically, no. It's not constitutionally required.
Practiacally speaking, you need to have either:
A) Won a major war
B) Served as governor of a state
C) Been in the senate
There aren't a hell of a lot of exceptions to this. People just won't take you seriously as a presidential candidate if you haven't done any of those.
