true or not, how did they sequence it to come to those conclusions without a sample?

The first published genome sequence was provided by the Chinese Government and was obtained from the lab at Wuhan. They had a sample. (What a surprise!)

The sequence has since been frequently updated by other independent researchers. That is how they know about the endless stream of variants (Delta, Lambda, Epsilon, etc.)

It is not technically difficult to isolate the COVID-19 virus and there no shortage of patients now from which to obtain samples.

There are still virologists who maintain that the variants of COVID-19 all contain sequences which bear markers as "gain-of-function" inserts which could have only occurred by manipulation in a laboratory settings. This is not a claim which the US Government is willing to publicly accept.
