I'm not sure what the black fleck is. I'm assuming it is part of the fiber itself since the orange goes beyond it and seems to encase it. The upper fiber does not have it though does it? Also the light line thru the one fiber is very distinct I would think of that type of fiber, however the other fiber DOES not show it. I'm thinking the colored background was used to hide the light line in the center of the one fiber that also has black specks.
