Since the District of Columbia is defined as the Federal preserve in the Constitution, wouldn’t changing that status from Federal seat of government to a State require an amendment to the Constitution?

Unconstitutional. The federal district was created precisely so as NOT to be under the jurisdiction of ANY state. This concept is easy to get. I got it in high school.

The ‘power-brokers’ of DC don’t want a Maryland-connection. They want an entire state government which they’d run.
I would add this...having lived around the region. As corrupted as Montgomery and PG counties are in Maryland...once DC is a state, it’ll probably triple the amount of corruption that you’d typically see. Whoever ends up as Governor...won’t last more than four years before a corruption case takes them down.
If we get back into power, perhaps we should consider dividing Wyoming into twelve different states.
While that would provide the representation that they say they are seeking, it would not yield the additional 2 D senators that they so desparately want.
