Not possible, even if he wanted to, all highways are owned by the states, not the Federal government.
The states get money from the Congress to build and maintain them, but the states own them.
Route numbering does not effect ownership.

It would surprise me if even half know the general distribution chain, let alone some of the specific “choke points”. Having been in agriculture/horticulture for the past 25yrs (retired Farmer), I’ve met a lot of astoundingly “unaware” people.
Around here, there are massive warehouse supermarket dry-distribution centers, similar to this one in FL, but once the roads are closed, the stores etc run out quickly.

Okay, let him shut down the Interstates. There are plenty of state highways that serve as alternate routes, which he can’t touch without declaring martial law. Deliveries will be slowed, but they would continue.
