They cannot run on same ticket! US law.
Also, In Florida you cannot run for POTUS and Senator at the same time. Florida law.

They can both run. The Constitution prohibits the electors from their home state from voting for both of them, so Rubio would lose the Florida electoral votes and possibly push the VP vote to the Senate if it had been close. Or Rubio could decide that he wants to live in Wyoming like Cheney did before he ran for VP.
